Introduction and Objectives: Chenopodium ambrosioides is an herbaceous shrub commonly known in Brazil as ‘mastruz’, a plant rich in flavonoids and terpenes, compounds that have diverse pharmacological properties including antioxidant and chemo-preventive agent for cancer , immunoestimulating property, anti-helmint action, genotoxic in vitro, anti-tumor activity and leishmanicidal effects. The aim of this study is to investigate the effect of hydroalcoholic extract of the leaves of C. ambrosioides treatment on models of inflammation and its action on the cell migration.
Methods and Results: Swiss mice males (2 months old, 25g) were orally treated, with 100 μL EBH at the doses of 5mg/kg (EBH5), 50 mg / kg (EBH50), or water apiogenic (Control) (n = 5), 1 / 1h for 6 hours for footpad oedema model induced by the injection of 50 μL of carrageenan 1%, for 6 or 17 consecutive days for granuloma model induced by the deployment of a cotton pellet in the back of the animal. The animals of 17 days of treatment were used to examine the weight of their organs and spleen, mesenteric and inguinal lymph node cellularities. The treatment with EBH 5mg/kg dose induced a reduction in the swelling at the 3rd (26.55%) and 24th hour (53.95%). For granuloma of 6 days, the groups EBH5 and EBH50 showed an increase of inflammatory liquid volume (EBH5: 13.72%; EBH50: 13.23%), while it induced inhibition of cell recruitment (EBH5: 39.03%; EBH50: 46.35%). Furthermore, the treatment of 17 days induced inhibition of both the footpad oedema (27.11%) and recruitment cell (39.14%) only in EBH5 group. While the spleen weight increased for EBH5 groups (34.05%) and EBH50 (77.51%), and mesenteric lymph node one for the group EBH5 (92.8%). Regarding the cellularity, there was no significant decrease of the spleen cell number for EBH5 (15.23%). As for EBH50 group, it was noted an increase of bone marrow (2.65%) and mesenteric lymph node (34.92%), and a reduction of inguinal lymph node(33.3%).
Conclusion: EBH Chenopodium ambrosioides treatment provides an antiedematogenic effect in acute inflammation and sub-chronic one at short treatments. As for cell migration, in sub-chronic inflammation at short treatments, there is an inhibitory effect, while there is, at prolonged treatment, on both cell migration and swelling, an inhibition one, even with an increase of some lymphoid organs cellularities. This shows a possible anti-inflammatory effect of the extract, but there is a need for further investigation.
